March 20, 2025 — Paris – Global biopharmaceutical giant Sanofi has announced its acquisition of DR-0201, a promising bispecific antibody developed by California-based biotech Dren Bio, in a deal valued at $600 million upfront and up to $1.9 billion in total. This acquisition positions Sanofi at the forefront of next-generation treatments for autoimmune diseases, with the potential to reset the immune system in patients with refractory conditions such as lupus.
What is DR-0201?
DR-0201 is a CD20-directed bispecific myeloid cell engager (MCE) designed to induce deep B-cell depletion through targeted phagocytosis. Unlike traditional B-cell therapies, which may struggle with incomplete depletion or treatment resistance, DR-0201 engages tissue-resident and circulating myeloid cells to eliminate pathogenic B cells at their root.
Currently in Phase 1 clinical trials, DR-0201 has shown encouraging results in both preclinical and early clinical settings—demonstrating robust, durable B-cell clearance. This mode of action holds potential not only for more effective symptom control but also for inducing treatment-free remission in autoimmune conditions.
A Strategic Leap in Immunology
Sanofi’s R&D chief Dr. Houman Ashrafian called DR-0201 “at the frontier of treating autoimmune diseases,” emphasizing its role in delivering breakthrough medicines where unmet medical needs persist.
“Through our own research and strategic licensing and acquisitions, we continue to advance our goal of becoming the leader in immunology,” said Ashrafian.
The acquisition reflects Sanofi’s continued pivot toward high-impact, immune-modulating therapies, a space that has seen explosive growth driven by biologics and precision immunotherapies.
Deal Details
Under the terms of the agreement:
- Sanofi will acquire DR-0201 via a dedicated affiliate, Dren-0201, for $600 million upfront
- Additional milestone payments could total $1.3 billion, linked to clinical and commercial achievements
- Dren Bio will retain its independence and continue advancing its broader pipeline of antibody therapeutics
The transaction is expected to close in Q2 2025, pending regulatory approvals.
